I was cleaning the dirt off of my mains today and found a soft spot on the main. I cleaned with purified water and toothbrush then put a fan and sulfur on it. Is this the best way or do you guys have other methods?

You need to cut out rotted portion or it will continue to spread.

That’s what the tooth brush does it cleans off all rot and only leave live tissue.

Cut the rot FULLLLLY and 100% daconil direct on it or you wont have a main in about a week

After doing the above, I'd consider exposing that area by removing the 1 or 2 leaves over it so condensation does not build up around it in the mornings. Let the sun dry it up along with a fan. But, you'll also need to have something over it when it rains. Rot spots even when fixed are a season long worry.

I'd do what Spence said,,,, he's the master of rot. I've had sections of rot on vines before and sprinkled straight clearys very lightly on the areas with good luck.

Straight Daconil is good on rot. Once dry it is harder for rain to wash it off. I often paint it on stems later in the season. It takes a good scrub to get it all off before weigh-off.
