Is one male flower enough? I usually have two males from one vine to pollinate a female, but lately I've had only one. I can always use two males from two vines, but that shoots the genetics. Ideas?

Theres plenty of pollen in one flower to have success.

Absolutely enough in one. It's a much more secure feeling to use multiple but if a specific cross is a priority, there is your deciding factor.
